From my experience, the Collector is periodically making outbound HTTPS connections to checkin. When it checks in, it looks for things like Collector updates, remote connection requests, Datasource updates, etc.
This is how most modern tools like LogicMonitor work with SaaS based items as a persistent tunnel flags a lot of items now adays.

The collector periodically checks in and downloads the updated collector "feed" if there are new tasks or changes. Whenever you do some action in portal that affects a collector, it updates the feed and then the collector checks in and reacts accordingly.

I don't really know the communications between the Collector and the platform, but I think of the Collector just frequently checking into the platform asking for any changes, scheduling, uploading new data, or to run new commands. But I don't really know myself if it's via reverse tunneling or just frequent polling. It kinda just works. I guess you might be able to sniff the traffic. It is encrypted and the key is in a config file, but the key rotates daily.
